MicroRNA-mediated incoherent feedforward motifs are robust

چکیده
In this paper, we integrate transcriptional and posttranscriptional regulation into microRNAmediated incoherent feedforward motifs and analyze their dynamical behaviors and functions. The analysis show that the behaviors of the system are almost uninfluenced by the varying input in certain ranges and the introducing of delay and noise. The results indicate that microRNA-mediated incoherent feedforward motifs greatly enhance the robustness of gene regulation.

The Role of Incoherent MicroRNA-Mediated Feedforward Loops in Noise Buffering
MicroRNAs are endogenous non-coding RNAs which negatively regulate the expression of protein-coding genes in plants and animals. They are known to play an important role in several biological processes and, together with transcription factors, form a complex and highly interconnected regulatory network. Looking at the structure of this network, it is possible to recognize a few overrepresented ...
متن کاملIncoherent feedforward motifs as immune change detectors
We speculate that incoherent feedforward loops may be phenomenologically involved in self/nonself discrimination in immune-infection and immune-tumor interactions, acting as “change detectors”. In turn, this may result in logarithmic sensing (Weber phenomenon) and even scale invariance (fold-change detection).
متن کاملCircuit Designing Robustness to Temperature in a Feedforward Loop
— 'Incoherent feedforward loops' represent important biomolecular circuit elements capable of a rich set of dynamic behavior including adaptation and pulsed responses. Temperature can modulate some of these properties through its effect on the underlying reaction rate parameters. It is generally unclear how to design such a circuit where the properties are robust to variations in temperature. H...
متن کاملFundamental Dynamic Units: Feedforward Networks and Adjustable Gates
Background: The activation/repression of a given gene is typically regulated by multiple transcription factors (TFs) that bind at the gene regulatory region and recruit RNA polymerase (RNAP). The interactions between the promoter region and TFs and between different TFs specify the dynamic responses of the gene under different physiological conditions. Results: By choosing specific regulatory i...
متن کاملSynthetic incoherent feedforward circuits show adaptation to the amount of their genetic template
Natural and synthetic biological networks must function reliably in the face of fluctuating stoichiometry of their molecular components. These fluctuations are caused in part by changes in relative expression efficiency and the DNA template amount of the network-coding genes. Gene product levels could potentially be decoupled from these changes via built-in adaptation mechanisms, thereby boosti...
